Imaging cathepsins: from cellular processes to in vivo diagnosis

Invited lecture, where different approaches for imaging cysteine cathepsins were presented. These included small molecule fluorescent activity-based probes, targeted liposomes and also DARPins.

Targeting of cathepsin S with stefin A conjugated liposomes in inflammation

In this Master thesis, a stefin A Asp61Cys mutant was prapared and found to inhibit both mouse and human cathepsins S and B. In addition, liposomes, conjugated to this stefin A mutant were prepared and their binding to cathepsins S and L confirmed in vitro. Finally, such liposomes were found to enter U87 cells, suggesting that the system may also work in cells.

Characterizatin of potent cathepsin B inhibitors selected from DARPin libraries

In this short lecture selection and characterization of two most promissing cathepsin B inhibiting DARPins was presented, including binding kinetics to human cathepsins and preliminary structural data for one of them.
