The aim of the study was to compare six commercial, three non-commercial, and two combined methods for isolation of DNA from milk. According to the real-time PCR efficiency (E) and correlation coefficient (R2), only three commercial kits, namely, QIAprep Spin Miniprep (Qiagen), DNeasy Blood and Tissue kit, (Qiagen) and SmartHelix First DNAid (ExVivon, Ljubljana, Slovenia), as well as one combined method (phenol-chloroform protocol and Nucleospin Food kit (Macherey-Nagel, Düren, Germany)produced DNA of good quality for real-time PCR. Given the estimated time required for DNA extraction, and the cost and labour requirements, the three commercial kits were faster, cheaper and simpler than the other methods, and can be recommended as applicable methods for the extraction of DNA from milk.

Potvorbe ovčjega (Ovis aries) in kozjega mleka (Capra hircus) in mlečnih proizvodov s cenejšim kravjim mlekom (Bos taurus) se najpogosteje pojavljajo zaradi pridobitve dodatnega dobička. Z namenom skrajšanja in poenostavitve kontrole takih nedovoljenih posegov v živila smo vpeljali PCR v realnem času kot metodo kvalitativnega in kvantitativnega določanja kravjega mleka v mleku in mlečnih proizvodih iz drugih vrst mleka.

Horizontal and vertical variations in the distribution of radiocarbon, d13C and d18O in groundwater of the Žitný island (Rye island) at the Danube River Basin, which is the largest groundwater reservoir in the Central Europe (about 10 Gm3), has been carried out using the geostatistical analysis of experimental isotope data. The core of the subsurface C14 profile represents contemporary groundwater with C14 values above 80 pMC, except a small area on the east of the island where C14 values down to 40 pMC were observed, indicating a groundwater source outside of the direct Danube River influence. This has been for the first time when vertical distributions of isotopes in different groundwater horizons have been studied. This research is important for determination of the sources of milk and also for determination of authenticity of milk and its adulteration with water.

The spatial variability of the mineral composition of grapevines in production vineyards along the east Adriatic coastwas determined and compared between conventional and sustainable vineyard management. Factor analysis reveals three factors with strong loading for the macronutrients K and P and the micronutrient Mn,which explain 67% of the total variance in the mineral composition. Here, the major contribution of these three elements can be explained by abiotic and biotic soil parameters, with soil concentrations of K, Fe and Cu, organic matter content etc. In addition, analysis of the mineral composition data shows significant differences between differently managed vineyards,with increased bioaccumulation of P and K in sustainable vineyards, while Zn bioaccumulation was increased in conventional vineyards. The transport of elements from soil to milk is important for geographical determination of milk.
