We have developed a 3-D in vitro model of CHO-K1 cells incorporated in collagen gel. We have determined optimal pulsing protocols and other parameters for sucesfull electrotransfection in such 3-D model. In parallel we have also determined efficency of electropermabilization for small molecules. Our results are comparable to in vivo results which enables us to optimize protocols in a more realistic enviroment. We have compared to types of pDNA administration, our results will enable us to develop a theoretical model pf pDNA mobility in collagen gel an extracellular matrix.
