Comparison of various spectroscopic imaging techniques for investigation of Hg and Se metabolism in plant tissues

This presentation focused on the development/application of imaging of the spatial distribution of Hg and Se in plants at the macro (organ/tissue) and micro (tissue/cellular) level. For elemental imaging both laser ablation-ICPMS and synchrotron radiation-based micro-X-ray fluorescence spectrometry techniques were used for fast, low-resolution () 8 μm) mapping of large areas (≥ 1 mm2) and high-resolution (sub-μm) mapping of small areas (≤ 0.05 mm2), respectively.

Studies of trace element homeostasis in plants using different imaging techniques

In order to be able to develop new biofortification technologies, and prevent accumulation of unwanted trace elements in plant food and food related products, extensive knowledge on the mechanisms involved in processes of trace element uptake, transport, accumulation, distribution and compartmentalization in plant tissues is needed. For this type of studies the use of techniques such as micro-XRF, micro-XAS, micro-PIXE and LA-ICPMS that enable imaging of the element distribution and speciation in plant tissues and cells with high spatial resolution and was presented together with the results of studies on Fe and Se distribution and speciation in different wheat genotypes and maize for the purpose of biofortification of selected crops.
