TURK, Boris. Lysosomal proteases as mediators or amplifiers of cell death?

An overview of protease signaling pathway in apoptosis was given with the emphasis on protease substrate identification. The results demonstrating that BCl-2 homologs are critical cysteine cathepsin substrates in apoptosis mediated by lysosomotropic detergents were also presented. In addition, the idea that lysosomes are destabilized downstream of mitochondria upon TNF-alpha stimulation, thereby potentially acting as signal amplifiers, was also presented.
