Control by pulse parameters of DNA electrotransfer into solid tumors in mice.

Electrically mediated gene transfer is dependent on electrical parameters used for tissue electroporation. The study evaluated efficacy of different electrical parameters in electrogene transfer and indicates that the best transfection efficacy is obtained using several 600 V/cm electric pulses duration of 5 ms.

Radiosensitizing effect of electrochemotherapy in a fractionated radiation regimen in murine tumors with different radiosensitivity

Electrochemotherapy uses cytostatics bleomycin or cisplatin that have radiosensitizing effect. Electroporation increses intracellular concentration of both drugs resulting in increased radiosensitization of tumors. The study has indicated as the first that electrochemotherapy is effective also in fractionated radiation regime, both on radisensitive and radioresistant tumors.

Controlled systemic release of interleukin-12 after gene electrotransfer to muscle for cancer gene therapy combined with ionizing radiation

Gene therapy with cytokine coding DNA is effective tumor treatment. For systemic transgene release the musle is suitable transfection site, enabling the controlled release. The study has demosntrated that this approch has good antitumor effect on local tumor growth and metastases, also in combination with tumor or metastatses irradiation.

Local and systemic antitumor effect of intratumoral and peritumoral IL-12 electrogene therapy on murine sarcoma.

In this study, we evaluated the antitumor effect of IL-12 electrogene therapy on murine SA-1 sarcoma. The IL-12 plasmid was applied either intra- or peritumorally. Intratumoral EGT resulted in 90% complete response rate to treatment with 60% resistance to challenge with SA-1 tumor cells. Peritumoral EGT resulted in16% complete response rate. Significant growth delay of untreated tumors at a distant site was achieved. These data suggest that IL-12 EGT may be useful in the treatment of soft tissue sarcomas.

Limb sparing treatment of bleeding melanoma recurrence by electrochemotherapy.

Bleeding metastases pose a serious clinical problem. In the study we have described a clinical case of bleeding metastases treated by electrochemotherapy. Single treatment stopped bleeding and had good antitumor effect.
