Speciation of arsenic in blood samples and cancerous cell structures with emphasis on analytical artefacts

On average the sum of arsenic species in blood plasma/serum of acute promielocyte leukaemia and multiple myeloma patients treated with arsenic is roughly 20% lower from total arsenic concentration in the same samples. In a case of blood cells the difference is even bigger indicating a large pool of "unvisible" arsenic in patient's body, which can only be seen with invasive methods including total digestion of samples.

Interactions of metalothioneins with metals in human astrocytoma cells

MTs are stress proteins involved in multiple resistance mechanisms of cancer cells. Their presence, inducibility and metal interactions were followed in glioblastoma cell line (U87) after exposure to arsenic trioxide. The results showed clear MT increase, Cu increase and low levels of arsenic imobilisation by MT.

Cytogenetic abnormalities in 40 Slovenian multiple myeloma patients

In this work we presented results on new genetic rearangements at 40 Slovene multiple myeloma patients. Since we only studied a small number of patients some rare rearangements were not found jet but we noticed coexistance of other rearangements. The research was ment as starting point for selection of patients for further tests based on preliminary research.

Determination of gene expression in human glia cell lines

Interactions of Cd with metalothioneins and its binding to protein was studied by monitoring of gene expression of metalothionein isoforms in human astrocitoma cell line (U87).Cells were treated with 1µM Cd (CdCl2) for different lenght of time (0 to 72 h). A real-time q-PCR was used to study gene expression for different MT isoforms ( MT1A, MT2A, MT3, MT1E, MT1X). GAPDH, TfR and Beta-aktin were used as reference gens. The work served as a control for As2O3 (bio)transformation in cells and its influence on induction of synthesis of selected MT isoforms.

Transformation of As2O3 in cell culture studies

The study was performed on human astrocytoma (glioblastoma) cell line U87 MG treated with 0.6, 2 and 6 microM arsenic for 12, 24 or 48 h. The gene expression for different MT isoforms (MT2A; MT1A, MT1F, MT1X, MT1E) was examined by measuring the mRNA levels with real time qPCR using SYBR Green I Master (Roche) for detection. GAPDH, Beta Actin and 18S rRNA were used as reference genes. At different conditions we noticed the increase or decrease of gene expression, above all in isoforms MT1X, MT1F, MT2A and MT3.
