The RT real-time PCR reaction was developed for the detection of Hop latent viroid (HLVd). The method was used for testing of hop samples known to be infected with the viroid. Standard curve was established and reaction parameters were set. No nonspecific reactions with other viroids were observed.

Grapevine yellow speckle viroid 1 (GYSVd-1) and Hop stunt viroid (HSVd) are most widespread viroids in grapevine. The RT real-time PCR was developed for the detection of both viroids in grapevine. Samples collected in different slovenian vineyards were tested for the presence of both viroids. Many infected samples were detected and mixed infections were frequently found. Standard curve using one of the positive samples was established and reaction parameters were set. No cross reactions with other viroids were observed.

Member of program commitee

The congress of International working group of legume and vegetable viruses was held in Ljubljana in August 2008. The most important researchers from the field participated on it. As a member of programm commitee I was involved in preparation of congress programe and selection of oral presentations. I also organized a field trip to show the participants the vegetable production in Slovenia and virus diseases we deal with.

TOPLAK, Nataša, KOVAČ, Minka. Molecula methods for fast detection and diagnosis of bioagents in drinking water and food

Molecular techniques can improve diagnosis in clinical laboratories. This review focuses on currently used molecular diagnostic methods. They improve sensitivity and decrease time needed for pathogen identification, they are used for determination of drug sensitivity. Currently most used techniques are restriction fragment length polymorphism, pulse-field gel electrophoresis, blot methods (Northern blot, Southern blot, Western blot and Dot blot), sequencing, polymerase chain reaction and variation like real time polymerase chain reaction (real-time PCR).
