The influence of genetic polymorphisms of GSTP1 on the development of asbestosis

Genetic factors play an important role in the development of asbestosis. A large superfamily of enzymes - glutathione S-transferases (GSTs) participates in detoxification of xenobiotics and toxic products of reactive oxygen species (ROS). In this research paper we were the first to show that GSTP1 genotype coding for an enzyme with high catalytic activity significantly increases the risk of developing asbestosis. This paper was already cited in the most comprehensive review paper on the current knowledge of genetic factors and the risk for asbestosis.

The influence of catalase genetic polymorphism on the development of asbestosis

Asbestos fibers trigger the production of reactive oxygen species (ROS) involved in the pathogenesis of asbestosis. As catalase (CAT) represents an important part of the enzymatic defense against ROS, we investigated if CAT-262C>T genetic polymorphism influences the risk of asbestosis. We observed a slightly elevated risk of asbestosis that increased after the involvement of cumulative asbestos exposure in subjects with the CAT-262TT genotype. These results contribute to understanding of interactions between genetic and environmental factors that may modify the risk of asbestosis.
