Primary metabolism deterines growth characteristics of all organisms as well as productivity of comercial organisms. A large pool of biochemical information has accumulated about the primary metabolism enzymes fro various organisms. These data can be used for metabolic engineering of primary metabolism pathways in order to enhance productivity of different industrial microorganisms.

LEGIŠA, Matic, MATTEY, Michael. Changes in primary metabolism leading to citric acid overflow in Aspergillus niger.

Strong anaplerotic reactions are characteristic for Aspergillus niger metabolism during accumulation of citric acid. The paper describes specific characteristics of A. niger regulatory enzymes that enable such conditions. The phenomenon of posttranslational modification of PFK1 enzyme is stressed as the most important.

ŠOLAR, Tina, TURŠIČ, Janja, LEGIŠA, Matic. The role of glucosamine-6-phosphate deaminase at the early stages of Aspergillus niger growth.

The paper describes glucosamine-6-phosphate deaminase characteristics which competes with 6-phosphofructo-1-kinase for identical substrate fructose-6-phosphate in Aspergillus niger cells. We have shown that deaminase from A. niger possesses specific kinetic features in respect to the enzymes form other organisms, enabling fast formation of glucosamine. Enzyme activity therefore decreases metabolic flux through glycolysis at the early stages of A. niger growth.

It has been shown that a highly active, citrate inhibition resistant form of PFK1 enzyme can be synthesised from a modified pfkA gene in A. niger cells. Normally an active shorter PFK1 fragment is formed after proteolytic cleavage of the C-terminal part of the native enzyme. After the insertion of modified genes into A. niger cells, increased citric acid productivity was recorded by transformants.

A patent application has been filed to protect the use of the modified mt-pfkA gene encoding a highly active shorter PFK1 fragment. A gene of microbial origine can cause deregulated metabolic flux in recipient microorganisms which is similar to that found in metastatic tumor cells. Deregulated glycolytic flux which acts as a strong anaplerotic reaction might enhance the synthesis of various metabolites of industial importance by commercial microorganisms.
