We performed a series of in vitro studies. Among other we showed that there is no direct correlation between increase of Mg2+ ions and transfection efficiency. This is in contrast with some other studies and can be explained by the effect of Mg2+ on stability of DNA in the cytoplasm and its binding to the cell membrane. We also found increased permeabilization of the cells with increased Mg2+ ions, which shows that the mechanisms of transfer for electrotransfection are different from transfer of small molecules.

COBISS.SI-ID: 7031636We related existing theoretical studies of electroporation with experimental results from planar lipid bilayers and cell lines in vitro. Measurements of conductivity before and after electroporation and correlation with permeability were also theoretically evaluated. Our analysis of the transmembrane voltage in cell suspensions and tissues and estimation of the diffusion are important for both analysis of intake of small molecules and intake of macromolecules like DNA with electrical pulses.
