Genetics of Streptomyces rimosus, the oxytetracycline producer

Paper summarizes research and industrial efforts in the area of molecular genetics of medically and industrially important oxytetracycline producer S. rimosus carried out over three decades. This article describes the procedures for in vivo and in vitro genetic manipulations in S. rimosus and their application to the study of the genetic instability of S. rimosus strains and the molecular cloning and characterization of genes involved in oxytetracycline biosynthesis. Novel compounds were generated using a biosynthetic engineering approach.

Total food chain safety : how good practices can contribute?

This is a review article about food quality and food safety, which have become a hot topic. “Food safety” is a broader term, which means an assurance that food will not cause harm to the consumer when it is prepared and/or eaten according to its intended use. Food safety point of view should be focused on knowledge, constant education and exchange of information. “From Farm to Table” approach is a philosophy with an important goal: safe and healthy food for all consumers. Motional restrictions of membrane proteins : a site-directed spin labeling study.

The method is based on site specific spin labelling of cysteine mutants of major coat protein of bacteriophage M13 reconstituted in lipid bilayers. We have used local constructs provided by neighbouring amino acids and lipid bilayer to obtain motional restriction on spin label along the primary structure of the protein. The work is an important step in the direction of membrane protein structure determination with SDSL-ESR method, which is becoming a popular alternative to high resolution techniques for membrane protein structure determination, due to their severe technical limitations.

Survival and stress induced expression of groEL and rpoD of Campylobacter jejuni from different growth phases

The article was published among top 5 % group of food microbiology journals. It is one of the first stress response research of campyloabacters on physiological level (protein synthesis, cross-resistance) and molecular level of gene expression. We described rapid and quantitative method for stress response detection on different cell level, which allow us to understand the response under stress combinations and stress resistant mechanism in cell populations.

Iron enriched yeast biomass - a promising mineral feed supplement.

Yeast biomass enriched with iron could represent a new and safer solution for prevention from anaemia development. An iron source is less toxic and has better absorbability in organisms. The purpose of our research was the determination of the most suitable ironsource in the cultivation medium for the yeast Saccharomyces cerevisiae. Iron(III) citrate, iron(III) chloride, iron(III) nitrate and Fe–EDTA complex were used. The uptake of the chosen iron compound, Fe(III) citrate, by the yeasts Candida intermedia and Kluyveromyces marxianus was also investigated.
