Abstract
Fresh and sea water organisms (cianobacteria, coelenterates, marine sponges) are screened for toxins using bio-assays, and cholinesterase inhibitors employing enzyme tests. In fresh water cianobacterial blooms, microcystins are determined and their levels correlated to ecological factors and abundance of photosynthetic algae. Mechanism of action of a wild-type and recombinant proteinaceous pore-forming toxin from the sea anemone Actinia equina and its mutants are studied using cells and artificial lipid membranes. Topology and kinetics of the toxin binding, insertion and pore formation are studied. Surface active and anticholinesterase compounds are isolated from marine sponges. Mechanism of anticholinesterase activity of 3-alkylpyridinium polymers isolated from the marine sponge Reniera sarai is studied using recombinant insect acetylcholinesterase. Their surfactant-like activity, e.g. hemolysis, is characterized as compared to that of low molecular surfactants.
