Significance for science
Discovery of new mechanisms of inhibition of TLR and their improvement is important for understanding the molecular mechanisms of cell signaling . The discovery of new details in the mechanism of cell signaling through TLR receptors and MyD88 adapter is important for understanding the function of innate immunity. Based of the detection of the impact of the INT domain, we can explain the inhibition of Myd88S alternative transcript. Discovery of the action of dimeric TIR domains allows to improve inhibition of the TLR signaling and contributes to the understanding of interactions between the TIR domains of proteins of the TLR signaling pathway, which is important for a number of inflammatory diseases, such as infections or chronic inflammatory diseases. Both types of inhibition are potentially interesting for therapeutic inhibition of inflammatory diseases. The project introduced the principles of synthetic biology in the manipulation of the immune response which is expected to provide innovative approaches to the treatment of an immune response, particularly inflammation. Of particular importance to science is the discovery of a modular implementation of DNA binding proteins for preparation orthogonal genetic NOR gate.
Significance for the country
Inflammation is associated with almost all diseases, from microbial infections to chronic inflammatory diseases such as atherosclerosis, arthritis and cancer. New approaches to the treatment of inflammatory diseases could be important for improving health, and on the other hand, also for the development of biotechnology in Slovenia. The project has direct relevance to the society through the achievement of high level of science and training of highly qualified personnel which represents the best investment for the economic development. The development of inventive methods of synthetic immunology is from an economic standpoint a perfect base for business initiatives. We filed patent applications for three inventions, which may allowing either their licensing or starting a company to commercially exploit the results. The project involved young researchers and graduate students and thus contributed to increasing the number of skillfully trained and innovative personnel in the field of biotechnology.
