Abstract
For introductory clinical trials of anti-tumor therapy with our newly developed TNF analogs, a substance of superior quality is needed. Therefore, a thorough study and application of rational approaches of production, including genetic and protein engineering, is necessary. Our project involves study of protein modifications leading to simplified and more economic isolation procedures, simple immobilization on solid supports and new approaches of renaturation. Besides TNF analogs, soluble TNF receptor p55 and chaperon GroEL from E. coli will be used as model molecules.
